Alkaline phosphatase/nuclear fast red staining

pink是nuclei，dark blue是enterocyte；

Alcian blue/nuclear fast red staining

light blue是Goblet的分泌物，pink同样是nuclei；

单细胞是必须要做的

• 细胞系没有做单细胞的必要，除非是Perturb-seq；
• 其次就是Patient或Mouse derived Organoid了；
• 最后就是纯in vivo的Mouse，这个周期长门槛高；

L-WRN细胞系：

• Wnt3a - Wnt激活剂，stem cell
• R-spondin3 - 辅助剂，提高Wnt的敏感度
• Noggin - BMP抑制剂，防止分化

ER medium：

• EGF - promoting proliferation
• R-spondin3 - 辅助剂，提高Wnt的敏感度

什么是conditioned media？这里的条件是指引分化的条件。
